The OM contains phospholipids only in its inner leaflet, while its outer leaflet is composed of glycolipids, primarily lipopolysaccharide (LPS). LPS is infamous for causing endotoxic shock associated with septicemia caused by Gram-negative organisms. The human innate immune system is sensitive to this molecule because it serves as a reliable indicator of infection.

Proteins in the outer membrane of Gram-negative bacteria can be split into two groups: lipoproteins and β-barrel proteins. Lipoproteins are proteins with lipids attached that embed them in the inner part of the outer membrane, and they are not considered transmembrane proteins. For example, E. coli has about 100 lipoproteins in its outer membrane, but scientists are unsure what most of them do. On the other hand, almost all the transmembrane proteins that are part of the outer membrane have a specific shape called a β-barrel. These proteins are made up of β sheets arranged in a cylindrical shape like a barrel.

Peptidoglycan

Bacteria have a rigid exoskeleton, and this is due to the presence of peptidoglycan in their cell walls. This gives them a simpler structure than Gram-negative bacteria. Without the outer membrane (OM), Gram-positive bacteria have thicker peptidoglycan layers to withstand the turgor pressure exerted on the plasma membrane. These layers of peptidoglycan contain long polymers called teichoic acids, which make up over 60% of the Gram-positive cell wall mass. Wall teichoic acids are covalently attached to peptidoglycan, while lipoteichoic acids are anchored to membrane lipids. These polymers contribute significantly to the structure and function of the cell envelope. Gram-positive bacteria also have various proteins that decorate their surfaces, and unlike Gram-negative bacteria, these proteins do not have an outer membrane to contain them. Meanwhile, gram-positive bacteria have a thick layer of peptidoglycan and teichoic acid, produce enterotoxins and neurotoxins, and are susceptible to some antibiotics.
